“Characterizing patterns of genetic risk in a living EHR-associated resource: the Colorado Center for Personalized Medicine Biobank”

The Colorado Center for Personalized Medicine (CCPM) Biobank is one of the largest institutional programs of its kind in the country, linking genomic data with phenotypes from across the entire electronic health record. To date, CCPM has consented over 200,000 people across the Rocky Mountain Region and has genome-wide data on ~80,000. Here, I will describe the dual clinical and research missions of CCPM, and describe our characterization of overall patterns of genomic risk using a repository of over 600 polygenic scores. These scores illustrate new opportunities for personalized medicine approaches across a number of disease domains: cancer, cardiometabolic, and autoimmune, for example, yet have major limitations for underrepresented populations. We provide some recommendations based on current evidence, describe ongoing research strategies to address this issue, and identify some aspects of polygenic scoring that better ensure that genomic insights are available to all.
